P12 - Epigenetics of Lioid Overliad during Aging: The Crutial Role of Acetyl-CoA

Principal Hypothesis and Objectives

We hypothesize that the pro-aging effects of lipid overload are transduced through a causal molecular chain that involves (i) enhancement of the nucleo-cytosolic acetyl-CoA pool leading to (ii) hyperacetylation of distinct histone sites and thereby (iii) epigenetic reprogramming of the cell. Disruption of this chain by mutation of the crucial histone sites identified may abolish the detrimental effects of lipotoxicity.
